FOX04-DRI is a modified peptide designed to selectively induce apoptosis (programmed cell death) in senescent cells — cells that have stopped dividing but refuse to die, accumulating with age and secreting inflammatory factors. Published in a landmark 2017 Cell paper by researchers at Erasmus University Medical Center, it represents one of the most targeted senolytic approaches in longevity research.
FOX04-DRI works by disrupting the interaction between FOXO4 and p53 in senescent cells. Normally, FOXO4 sequesters p53 in the nucleus, preventing p53 from triggering apoptosis — which is how senescent cells evade programmed death. FOX04-DRI competes with endogenous FOXO4, releasing p53 to induce apoptosis selectively in senescent cells while leaving healthy cells unaffected.
Important: The evidence base for FOX04-DRI varies by application. Many findings are from preclinical (animal or in vitro) studies. Large-scale human clinical trials may not be completed. Always evaluate the quality of evidence before drawing conclusions.
Limited safety data exists beyond the original preclinical studies.
